IMAGING AND HISTOPATHOLOGY FACILITY

IMAGING AND HISTOPATHOLOGY FACILITY

The following imaging facilities exist in Dr. Nurul Kabir's Lab in room #P-134, PCMD, ICCBS, Karachi University. All systems are run by NIS-Elements software from Nikon.
1) TE 2000E microscope:Fully motorized inverted microscope capable of
a) Brightfield
b) DIC (differential interference contrast)
c) Multichannel fluorescence (UV-2A, G-2A, B-2A and Cy5 filter cubes)
d)TIRF (total internal reflection microscopy) at very high resolution (N.A. 1.49).

The microscope is also equipped with live-cell imaging, real-time deconvolution and 3D reconstruction abilities. It is set up on a vibration-isolated platform.
Digital camera: Nikon DS-5M (color), DS-2MBWc

2) 90i microscope: Fully motorized upright microscope capable of
a) Brightfield
b) DIC (differential interference contrast)
c) Multichannel fluorescence (DAPI, FITC, TxR, FITC-TxR double cube and DAPI-FITC-TxR triple cubes) microscopy at very high resolution. The microscope is also equipped with real-time deconvolution ability.

In addition we have an Olympus E330 digital SLR camera with polarizer, UV filter and macro lens.

 

Dr. Nurul Kabir’s cell biology laboratory is currently engaged in research involving development of various in-vitro and in-vivo disease models related to the national need of Pakistan. Immunocytochemistry and immunohistochemistry, especially in paraffin sections, is widely used in all the disease models. Currently the lab is engaged in research related to the following disease models: 1) Kidney-stone 2) Hepatitis 3) Duodenal ulcer 4) Diabetes 5) Cancer and others.

The following two pictures show triple-label fluorescence immunohistochemistry for the nucleus, glucagon and insulin along with DIC done on 6 micrometer paraffin sections of rat pancreas by Dr. Hafizur Rahman, Faculty, PCMD

As can be seen, insulin and glucagon is only present in the islet of Langerhans of the panceras. Triple and quadriple label merges are also shown below. In rats which were made diabetic by injecting streptozotocin, the islet of Langerhans was small with very few insulin-positive beta-cells whereas there was an increase in the number of glucagon- positive alpha-cells as shown below.
